目的 比较地黄乙醇提取物(醇提物,EE)及其药渣水提物(RWE)的抗抑郁作用。方法 雄性昆明小鼠随机分为对照组、阳性药盐酸氟西汀(FH,10 mg ·kg-1)、地黄乙醇提取物及地黄药渣水提物的低、高剂量(2, 4 g ·kg-1,以生药计),连续灌胃(ig)给药7 d(每天1次),末次给药后1 h,进行开场实验(OFT)、悬尾实验(TST)、强迫游泳实验(FST)和利血平拮抗实验(RRT)测试;采用HPLC测定地黄提取物中主要药效物质梓醇含量。结果 对照组小鼠在悬尾实验、强迫游泳实验的不动时间分别为(91.0±28.3)、(98.0±30.4)s,对照组在利血平拮抗实验的体温为(35.0±0.7)℃,地黄醇提物低剂量、高剂量(2, 4 g ·kg-1)、药渣水提物高剂量(4 g ·kg-1)和盐酸氟西汀(10 mg ·kg-1)均显著缩短了不动时间(P<0.05)、逆转了体温降低(P<0.05)。开场实验中,模型组小鼠3 min内穿格次数为(91.9±24.6)次,各给药组与之相比均无显著差异;地黄乙醇提取物中梓醇含量为33.48 mg ·g-1原药材,地黄药渣水提物中为0.034 mg ·g-1原药材。结论 地黄醇提物及其药渣水提物均有抗抑郁作用,其作用机制可能涉及单胺能神经系统。
Abstract
OBJECTIVE To investigate the antidepressant-like effect of extracts from dried root of Rehmannia glutinosa Libosch.(Dihuang)(RG). METHODS Mice were treated with RG ethanol extract(EE), the residue water extract(RWE) from EE, and fluoxetine hydrochloride(FH, 10 mg ·kg-1) everyday for seven consecutive days by intragastric administration(ig). Mouse forced swim test(FST), tail suspension test(TST), open field test(OFT) and reserpine reversal test(RRT) were used to observe antidepressant-like effects of RG extracts, respectively, at 1 h after the last administration. RESULTS The immobility time of TST and FST in model mice was(91.0 ± 28.3),(98.0 ± 30.4) s, respectively, and the body temperature in model mice was(35.0 ± 0.7) ℃. There was significant decrease among different ratios of EE(2, 4 g ·kg-1), RWE(4 g ·kg-1) and FH(10 mg ·kg-1) groups compared with model group. The numbers of crossing squares in 3 min in model mice during OFT were(91.9 ± 24.6), and there was no significant difference among EE(2, 4 g ·kg-1), RWE(2, 4 g ·kg-1) and FH(10 mg ·kg-1) groups compared with model group. The contents of catalpol in EE and RWE were determined for 33.48 and 0.034 mg ·g-1. CONCLUSION Both EE and RWE exert antidepressant-like effect, and the mechanism underlying may be involving monoaminergic nervous systems.
